"BACKGROUND: Strategies to discover circulating protein markers of ovarian cancer are urgently needed. We developed a novel technology that permits us to isolate recombinant antibodies directed against the potential serum biomarkers, to facilitate the further development of affinity reagents necessary to construct diagnostic tests.

Paper: Systematic evaluation of candidate blood markers for detecting ovarian cancer.

"BACKGROUND: Epithelial ovarian cancer is a significant cause of mortality both in the United States and worldwide, due largely to the high proportion of cases that present at a late stage, when survival is extremely poor. Early detection of epithelial ovarian cancer, and of the serous subtype in particular, is a promising strategy for saving lives. The low prevalence of ovarian cancer makes the development of an adequately sensitive and specific test based on blood markers very challenging.

Paper: Effect of normalization methods on biomarker identification utilizing mass spectrometry.

"In biomarker identification using mass spectrometry, normalization makes it possible to compare mass spectra obtained from different samples. However, the relative influence of different normalization methods is an unexplored topic. In this study, we compared the most widely used normalization methods in a systemic manner to investigate impact of normalization. According to our experimental results, different normalization methods result in the selection of different features of different discriminatory power."

To date, no blood or stool biomarkers with both high sensitivity and specificity for potentially curable early stage disease have been validated for clinical use. SELDI and MALDI profiling are being used increasingly to search for biomarkers in both blood and urine. Both techniques provide information predominantly on the low molecular weight proteome (<15 kDa).

"INTRODUCTION: We evaluated the ratio of free to total prostate-specific antigen (PSA) and PSA to protein concentrations in saliva and serum of healthy men. MATERIALS AND METHODS: Concentrations of protein, free PSA, and total PSA in serum and saliva were measured in 30 healthy men aged 42 to 73 years, and their ratios were compared between the two fluids. RESULTS: There was a significant direct correlation between serum free-total PSA ratios of serum and saliva (P = .04) and between total PSA-protein ratios of serum and saliva (P = .02).

"OBJECTIVE: To evaluate if serum levels of candidate ovarian cancer biomarkers vary with individual characteristics of healthy women who are likely candidates for an ovarian cancer screening program. METHODS: We analyzed serum CA125, mesothelin, and HE4 levels in a sample of 155 healthy postmenopausal women at increased risk for developing ovarian cancer based on personal and family cancer history. Information on reproductive, family and medical histories, lifestyle factors, and anthropometry was collected by self-report.

Paper: Prediction of ovarian cancer prognosis and response to chemotherapy by a serum-based multiparametric biomarker panel.

"Currently, there are no effective biomarkers for ovarian cancer prognosis or prediction of therapeutic response. The objective of this study was to examine a panel of 10 serum biochemical parameters for their ability to predict response to chemotherapy, progression and survival of ovarian cancer patients. Sera from ovarian cancer patients were collected prior and during chemotherapy and were analysed by enzyme-linked immunosorbent assay for CA125, kallikreins 5, 6, 7, 8, 10 and 11, B7-H4, regenerating protein IV and Spondin-2.

Paper: IPO-38 Is Identified as a Novel Serum Biomarker of Gastric Cancer Based on Clinical Proteomics Technology.

"Gastric cancer is one of the most common malignancies in China. So far, there are few reliable serum biomarkers for diagnosis. The available biomarkers of CEA, CA19-9 and CA72-4 are not sufficiently sensitive and specific for gastric cancer. In this study, a high density antibody microarray was used for identifying new biomarkers from serum samples of gastric cancer. Serum samples from colorectal cancer, pancreatic cancer, hepatocellular cancer, and breast cancer were also screened for comparative study. As result, some candidate biomarkers were identified.
